Telomeric impact of conventional chemotherapy

Abstract:

The increased level of chromosome instability in cancer cells, leading to aneuploidy and gross chromosomal rearrangements, is not only a driving force for oncogenesis but also can be the Achille’s heel of the disease since many chemotherapies (CT) kill cells by inducing a non-tolerable rate of DNA damage. A wealth of published evidence showed that telomere stability can be more affected than the bulk of the genome by several conventional antineoplasic drugs. These results raise the interesting possibility that CT with genotoxic drugs preferentially target telomeres. In agreement with this view, accelerated shortening of telomere length has been described in blood lineage cells following high-dose CT (stem cell transplantation) or non-myeloablative CT. However, almost nothing is known on the consequences of this shortening in terms of telomere stability, senescence and on the development of second cancers or post-treatment aging-like syndromes in cancer survivors (cognitive defect, fertility impairment, etc.). In this article, we propose: (1) telomeres of cancer cells are preferential genomic targets of chemotherapies altering chromosome maintenance; (2) telomere functional parameters can be a surrogate marker of chemotherapy sensitivity and toxicity; (3) the use of anti-telomere molecule could greatly enhance the sensitivity to standards chemotherapies.

Abstract: Dry yeast cells (DYC) were used as a cheap nitrogen source to replace expensive yeast extract (YE) for L-lactic acid production by thermophilic . Cassava starch (200 g·L ) was converted to L-lactic acid by simultaneous saccharification and fermentation using WCP10-4 at 50 °C in the presence of 20 g·L of DYC, giving 148.1 g·L of L-lactic acid at 27 h with a productivity of 5.5 g·L ·h and a yield of 92%. In contrast, 154.4 g·L of lactic acid was produced at 24 h with a productivity of 6.4 g·L ·h and a yield of 96% when equal amount of YE was used under the same conditions. Use of pre-autolyzed DYC at 50 °C for overnight slightly improved the lactic acid titer (154.5 g·L ) and productivity (7.7 g·L ·h ) but gave the same yield (96%).

Feasibility of Scaling up the Cost-Competitive and Clean Electrolytic Hydrogen Supply in China

Abstract: Scaling up clean hydrogen supply in the near future is critical to achieving China’s hydrogen development target. This study established an electrolytic hydrogen development mechanism considering the generation mix and operation optimization of power systems with access to hydrogen. Based on the incremental cost principle, we quantified the provincial and national clean hydrogen production cost performance levels in 2030. The results indicated that this mechanism could effectively reduce the production cost of clean hydrogen in most provinces, with a national average value of less than 2 USD·kg−1 at the 40-megaton hydrogen supply scale. Provincial cooperation via power transmission lines could further reduce the production cost to 1.72 USD·kg−1. However, performance is affected by the potential distribution of hydrogen demand. From the supply side, competitiveness of the mechanism is limited to clean hydrogen production, while from the demand side, it could help electrolytic hydrogen fulfil a more significant role. This study could provide a solution for the ambitious development of renewables and the hydrogen economy in China.

Effect of calcium lactate on compressive strength and self-healing of cracks in microbial concrete

Abstract: This paper presents the effect on compressive strength and self-healing capability of bacterial concrete with the addition of calcium lactate. Compared to normal concrete, bacterial concrete possesses higher durability and engineering concrete properties. The production of calcium carbonate in bacterial concrete is limited to the calcium content in cement. Hence calcium lactate is externally added to be an additional source of calcium in the concrete. The influence of this addition on compressive strength, self-healing capability of cracks is highlighted in this study. The bacterium used in the study is and was added to both spore powder form and culture form to the concrete. spore powder of 2 million cfu/g concentration with 0.5% cement was mixed to concrete. Calcium lactates with concentrations of 0.5%, 1.0%, 1.5%, 2.0%, and 2.5% of cement, was added to the concrete mixes to test the effect on properties of concrete. In other samples, cultured with a concentration of 1×10 cells/mL was mixed with concrete, to study the effect of bacteria in the cultured form on the properties of concrete. Cubes of 100 mm×100 mm×100 mm were used for the study. These cubes were tested after a curing period of 7, 14 and 28 d. A maximum of 12% increase in compressive strength was observed with the addition of 0.5% of calcium lactate in concrete. Scanning electron microscope and energy dispersive X-ray spectroscopy examination showed the formation of ettringite in pores; calcium silicate hydrates and calcite which made the concrete denser. A statistical technique was applied to analyze the experimental data of the compressive strengths of cementations materials. Response surface methodology was adopted for optimizing the experimental data. The regression equation was yielded by the application of response surface methodology relating response variables to input parameters. This method aids in predicting the experimental results accurately with an acceptable range of error. Findings of this investigation indicated the influence of added calcium lactate in bio-concrete which is quite impressive for improving the compressive strength and self-healing properties of concrete.

Abstract: China’s mega urban regions are focal points of economic development and environmental concerns. This paper positions four mega urban regions (i.e., the Yangtze River Delta, the Pearl River Delta, the Bohai Bay Area, and the South-eastern Fujian Province) along China’s coast into the national economy and elaborates their development challenges and planning innovations. Mega urban regions in China are spatial consequences of rapid economic transition. They deserve close scrutiny and demands for innovative planning responses in order to maintain their key role in driving economic growth but limiting their greenhouse gas emission.

Design of damping valve for vehicle hydro pneumatic suspension

Abstract: According to the design features of a hydro pneumatic spring, the necessity of a separate damping valve is proposed. Based on a 1/4 vehicle linear suspension model, the optimum damping coefficient is worked out and the parameters of the damping valve are determined with the equivalent linearization method. A practical structure of the damping valve is proposed having a small size, high flowrate when the valve opens, and the ability of enduring high back pressure. Based on bench tests, the damping valve has been found to properly work and be suitable. The design method and damping valve structure are useful guides for hydro pneumatic suspension, especially for the design of heavy-duty vehicles.
